Earl Sutherland discovered cyclic AMP (cAMP) as the first intracellular second messenger, showing that hormones act not directly on cellular machinery but through an intermediary molecule produced at the membrane. The finding established the cellular signaling cascade paradigm and provided the functional measurement point that enabled Lefkowitz's experimental demonstration of membrane receptors a decade later.
